Drug Category: Cardiotonic Agents/ Respiratory Smooth Muscle Relaxants
Indication: For the treatment of bronchospasm due to asthma, emphysema and chronic bronchitis.
Composition:
Each 5ml contains:
- Aminophylline………………………32mg
- Ammonium Chloride………………..30mg
- Diphenhydramine HCl……………….8mg
- Menthol……………………………0.98mg
Pharmacological Action:
- Aminophylline: Theophylline is structurally related to theobromine and caffeine. The precise mechanism of action of theophylline is not known, however, it is thought to be a phosphodiesterase inhibitor which may give a bronchodilatory effect. It also binds adenosine receptors.
- Ammonium Chloride: Ammonium Chloride is an electrolyte (substance that dissociates into ions when melted or dissolved in a suitable medium and thus allows electricity to pass). Ammonium Chloride is used in treatment of alkalosis (accumulation of base in blood and tissues). Ammonium Chloride is also used as expectorant and diuretic.
- Diphenhydramine: Diphenhydramine competes with free histamine for binding at HA-receptor sites. This antagonizes the effects of histamine on HA-receptors, leading to a reduction of the negative symptoms brought on by histamine HA-receptor binding.
- Menthol: Menthol primarily activates the cold-sensitive TRPM8 receptors in the skin. Menthol, after topical application, causes a feeling of coolness due to stimulation of ‘cold’ receptors by inhibiting Ca++ currents of neuronal membranes. It may also yield analgesic properties via kappa-opioid receptor agonism.
